This is a Ph.D. dissertation. Dementia is a major cause of disability and mortality. The most frequent type of dementia is Alzheimer's disease, accounting for about 70% of all dementia cases. Worldwide, about 20 million people suffer from Alzheimer's disease. With the increasing number of elderly in the world, Alzheimer's disease represents more and more a major health problem with an enormous impact on individuals, families, the health care system, and the society. Therefore, it is imperative that we understand the etiologies, the risk factors and the pathogenic mechanisms involved in Alzheimer's disease in order to develop effective treatments that ameliorate or even prevent this illness.
